endometrial biopsy to diagnose luteal phase defect

My doctor mentioned that 2 rounds of biopsying might be needed to determine if I have a luteal phase defect. Why is that? If the biopsy sample is of good quality, can it be determined with one biopsy? Guess I am a little bit in a hurry--am going on 41--and would like to proceed with trying again for a pregnancy, with supplementation if needed, ASAP. Probably a stupid question, but if a second biopsy is needed, do I definately need not to get pregnant in that cycle? Also, I understand that the local anesthetic is pretty uncomfortable---any truth to that rumor (I hope not:))?
