You may have a vaginal muscle spasm, irritation of the vagina, herpes, neuromuscular damage, or any of the number of possibilities. If you know where in the vagina, or the proximity of where in the vagina you have this pain, a biopsy might be indicated. The next time you have this pain I would see your gynecologist as soon as possible. No offense to family practice physicians, or certified registered nurse practitioners, but I would definitely see gynecologists. I have personally picked up to many abnormalities that really should have been picked up by others, including most recently an invasive vulvar cancer.
