To check if amniotic fluid is present a piece of litmus paper is used but the paper turns blue if it *is* amniotic fluid. The other test you are referring to is where the fluid is put onto a slide and when it dries it makes a fern-like pattern which can be seen under the microscope. Let's hope a doc jumps in to clarify this for us all. :)
