Dear Carl, There could be several reasons this could happen, ranging from using more concentrated urine/more sensitive test(1st test, which was done in the morning), followed by less sensitive tests and/or less concentrated urine(2nd test was in the afternoon)which might not register the presence of HCG early in pregnancy as it might take a week longer to get a positive result with a less sensitive test or weak sample. The Positive test may have detected a recent pregnancy or chemical pregnancy/miscarriage with declining levels of HCG. Some factors may interfere with tests causing false-positive or negative results. Is your wife on any medication Rx or OTC? You can repeat the test in a week with first am urine or see a doctor and get a blood test to confirm pregnancy or pursue any health issues. It's frustrating but I hope this helps. Marie
