No, the tests do not have to be on a certain day but when the results are read, you do have to know what day inyour cycle it was so that they can be accurate.

Example: LH

if you are in the follicular phase, the range is 5 - 15 mIU/ml.

If you are in the midcycle (ovulation) normal range is 30 - 60 mIU/ml.

If you are in the luteal phase the range goes back down to 5 - 15.
