The 530D has a pretty good vag probe.  My guess is that it was undectable the day before.  Remeber there must be a finite point in time when a GS is detectable. It has nothing to do with waiting 48, 72, or 96hours.; that has to do with the reproducibilty of an increasing quant beta.
