I read the stuff about a permanent menopause in the leaflet with the drug, it says something like "rarely, some women may not resume menses at cessation of the drug" and the pituituary gland stuff came from a web page called the national lupron victims network http://www.voicenet.com/~nlvn/menopaus.html. It says, I quote (the references are on the page and I think some of them have been misused as I looked at some of the articles, so I'm not sure what to believe) :

"DOWN-REGULATION or DESTRUCTION of PITUITARY RECEPTORS:

Downregulation of Receptors - "the unusually rapid loss of receptor activity called 'downregulation' has been been confused with receptor blockade [blocking the receptor] and is now understood to be due to a disappearance of the receptor." (13)

1. "Continuous therapy with leuprolide [Lupron] apparently produces a decrease in the number of pituitary GnRH receptors." (14) 2. Friedman et al stated that "down-regulation of pituitary GnRH receptors is usually achieved" "between treatment weeks 2 through 7." (15)

* LUPRON'S EFFECT ON THE PITUITARY: 1. When a GnRH analog was given to rats in a breast cancer study, after just one month there was a 21% reduction in the weight of the anterior pituitary. (27) 2. In 1990, Word et al stated that "there is evidence that GnRH-a may have direct effects in pituitary cells other than gonadotropes." He also stated that "little is known regarding the effect of GnRH-a on other aspects of pituitary function." (28) 3. In 1992, 7 years after the first approval of Lupron, Cedars et al stated that "no thorough evaluation of the effect of these substances [GnRH-a] on the pituitary, the site of supposed action, has been reported." (29) 4. Janaky et al stated that "agonistic and antagonistic analogues of LH-RH were also reported to be internalized in pituitary cells by endocytosis." (30) "
